Pam3CSK4 is a TLR 1/2 agonist and a cell-permeable, water-soluble synthetic tripalmitoylated lipopeptide that mimics the acylated amino terminus of bacterial LPs. Bacterial LPs are a family of pro-inflammatory cell wall components found in both Gram-positive and Gram-negative bacteria. Pam3CSK4 is a potent activator of the pro-inflammatory transcription factor NF-κB. It induces production of TNF-α, superoxide, NO, and IL-6, stimulates phosphorylation of p100/p110 and p60 in granulocytic-differentiated HL-60 cells, promotes differentiation of naive CD4+ T cells into Th17 cells, and enhances tyrosine protein phosphorylation and activation of ERK1/2 and MEK1/2. It is used in prevention of inflammation caused by certain bacterial infection.
